有资料显示有用银染色显示黏液的方法，是否真实？

文献中有过此类报道，但至今也未见有大的推广使用，但作为一种特定的技术手段，此处还是应做个介绍：

1.组织处理：

组织甲醛固定，常规脱水、透明、石蜡包埋，切片4μm。

2.染液配制：

（1）六亚甲四胺银贮备液：

①六亚甲四胺水溶液：六亚甲四胺3g+蒸馏水100ml。

②5%硝酸银水溶液：硝酸银5g+蒸馏水100ml。

③5%硼砂（四硼酸钠）水溶液：硼砂5g+蒸馏水100m l。

④六胺银工作液：

取3%六胺水溶液40ml加入5%硝酸银2ml即产生白色沉淀，振荡后即溶解而澄清，然后加入5%硼砂6ml，摇匀。此液临用时配制。

（2）0.5%高碘酸水溶液：

高碘酸0.5g+蒸馏水100ml。

（3）0.2%氯化金水溶液：

氯化金0.2g+蒸馏水100ml。

（4）3%硫代硫酸钠水溶液：

硫代硫酸钠3g+蒸馏水100ml。

3.染色方法：

（1）石蜡切片4～5μm，脱蜡至水洗。

（2）0.5%高碘酸氧化30分钟。

（3）自来水洗，再充分蒸馏水洗。

（4）入六胺银工作液于70～80℃温箱内约40～60分钟至切片变棕色（烟草色）。

（5）自来水冲洗。

（6）0.2%氯化金调色（直至切片棕色变灰褐色）。

（7）自来水洗，再蒸馏水洗。

（8）3%硫代硫酸钠水溶液10～20秒显色。（如对比染色可用VG或Masson）

（9）常规脱水，透明，封固。

4.结果判定：

染色结果显示胃黏膜上皮萎缩性胃炎肠上皮化生，肠黏液腺癌阳性。在含黏液的细胞胞质内乃至细胞核内均可见分布均匀的黑色银染颗粒，背景清晰，反差较强。5.此法优势：此染色法对显示胃、肠上皮黏液及宫颈腺黏液很有意义。六胺银液染色显示黏液敏感性高，颗粒清晰，质感强，当病理组织含黏液较少时，用此法可较清楚地显示，对癌组织的鉴别诊断很有裨益。可作为阿新蓝-PAS等，显示黏液方法的补充和对照。此染色法染液配制简单，操作步骤不复杂，易于掌握。

6.注意事项：

（1）配制银溶液的器皿要十分清洁，配制时的银溶液如有乳白色混浊，表示器皿不洁，应倒掉重新配制。

（2）六胺银液中的染色时间要根据切片的厚薄及温度而定，薄切片用时短，厚切片用时长温度低需时长（如50℃时需1～2小时）；温度高需时短（如80℃仅需40分钟）；最好是加温浸染30分钟后，显微镜下观察。

（3）氯化金调色，时间不能过长，否则切片色调暗红。
